Abstract:

Background: The aim of this study was evaluation of developmental rate and Bax, Bcl-2 and ErbB4 genes expression following re-vitrification in compact and early blastocysts stages. Materials and Methods: 5-8 cell embryos were collected from female mature mice, 60-62 hours post hCG injection. The embryos were divided to five groups including: fresh, vitrified at 5-8 cells, vitrified at blastocyst stage, vitrified 5-8 cells and re-vitrified at compact or early blastocyst stage. Embryos were vitrified by using cryolock, as described by Kuwayama et al. Development of embryos were recorder daily. Results were analyzed by using Chi square. QPCR has been used to examine the expression of Bax, Bcl2 ErbB4 genes in derived blastocysts. The data was analyzed with ANOVA. Results: Blastocyst formation rates showed no significant differences between vitrified and re-vitrified groups, although there was significant difference between re-vitrified and fresh embryos (p<0.05). Also it showed no significant differences between two re-vitrified groups. Our data showed that expression of Bax, Bcl- 2 and ErbB4 genes had a significant difference (p<0.05) between re-vitrified and fresh embryos. Although there was no significant difference between re-vitrified and vitrified embryos regarding genes expression. Expression of Bax, Bcl-2 and ErbB4 genes showed no significant difference between two re-vitrified groups. Conclusion: Based on our study, re-vitrified embryos in compact and blastocyst stage are nearly equal in developmental rate and expression of Bax, Bcl2 and ErbB4 genes, although this amount of genes expression is altered significantly compared with fresh embryos. Therefore, we can re-vitrify embryos in compact or blastocyst stage.

Which stage of mouse embryos is more appropriate for vitrification?

Objective Vitrification has been shown as one of the most effective methods of cryopreservation for mammalian embryos. However, there is no consensus which stage of embryonic development is the most appropriate for vitrification with subsequent maximal development after thawing. This study was carried out to explore and compare the effect(s) of vitrification on mouse 2-cell, 4-cell, 8-cell, mor...

P-30: Developmental Capacity and Blastocyst Formation of Thawed Tetrahedral Versus Non-Tetrahedral 4-cell Stage Mouse Embryos After Vitrification

Background: It was reported in a literature that approximately one third of the 4-cell stage embryos did not exhibit a tetrahedral shape. Non-tetrahedral embryos showed a lower in vitro developmental potential than tetrahedral embryos. Recently vitrification technology has been widely employed for embryo cryopreservation. The objective of this study was to prove our hypothesis that vitrified - ...

Effects of Re-Vitrification of Mouse Morula and Early Blastocyst Stages on Apoptotic Gene Expression and Developmental Potential

OBJECTIVES Re-vitrification of embryos immediately after thawing or after a culture period could be used to preserve the extra embryos after embryo transfer. This study aims to clarify the effect of re-vitrification on Bax and Bcl-2 gene expressions of compact and early blastocyst stage embryos. MATERIALS AND METHODS This experimental study was performed on mouse embryos. We collected 8-cell ...

Development of 4-cell mouse embryos after re-vitrification.

This paper reports studies on the effects of re-vitrification by the CPS (Closed Pulled Straw) method on the development of 4-cell stage mouse embryos. The procedure involved culturing 2-cell mouse embryos in G-1 medium until the 4-cell stage followed by the division of the normal 4-cell stage embryos into a control group (non-vitrified) and two experimental subgroups (vitrified and re-vitrifie...
